生物薄片免疫荧光检测抗核抗体的应用与评价 被引量:7

摘要 目的 评价生物薄片免疫荧光技术 (BIOCHIP IFA)检测抗核抗体 (ANA)在自身免疫性疾病诊断中的应用价值。方法 采用生物薄片IFA技术 ,使用 2种抗原基质 (猴肝 /Hep 2细胞 )检测 73例自身免疫性疾病患者和 2 0例正常人ANA ,并与纯化多种核抗原包被ELISA、常规以鼠肝为抗原基质的IFA法进行比较。结果 生物薄片IFA的阳性率为 93.2 % ,ELISA阳性率为 95 .9% ,常规以鼠肝为抗原基质的IFA阳性率为 81.9%。生物薄片IFA阳性率与ELISA比较差异无显著性 (P >0 .0 5 ) ,但与鼠肝为抗原基质的IFA比较差异有显著性(P <0 .0 5 )。结论 生物薄片IFA检测ANA具有标准化、直观、灵敏、快速的特点 ,对提高自身免疫性疾病的实验诊断水平及指导临床治疗有一定的实用价值。 Objective To evaluate the immunofluorescence assay(IFA) BIOCHIP reagent kit in the detection of antinuclear antibodies(ANA) in patients with autoimmune disease. Methods Serum samples were tested with the BIOCHIP reagent which contained two substrates: human epithelial cells(Hep-2) and primate liver. The results were compared with other two methods, one is ELISA, the other is routine IFA with mouse liver substrate. Results The positive rates of BIOCHIP IFA, ELISA and routine IFA are 93.2%, 95.9% and 81.9% respectively. There are no statistical difference between the results of BIOCHIP IFA and ELISA( P >0.05), but they all have significant differences( P <0.05) with routine IFA. Conclusions The BIOCHIP IFA is sensitive, quick and can be standardized. It is useful for clinical diagnosis of autoimmune diseases.
