摘要
利用醇脱氢酶(ADH)催化乙醇与氧化型烟酰胺腺嘌呤二核苷酸 (NAD)反应的原理 ,通过测定还原型烟酰胺腺嘌呤二核苷酸 (NADH)吸光度的变化率得出其酶促反应速度 ,对应不同的乙醇含量而制得标准曲线 ,试样中乙醇含量由测定值查标准曲线求得 ;讨论了pH值和抑制剂对测定的影响 ;方法简便、快速。
An enzymatic method was applied to determine ethanol based on the reaction of ethanol and nicotiˉ namide adenine dinucleotide in oxidized form(NAD)catalyzed by alcohol dehydrogenase(ADH).The change rate of the absorbance of nicotinamide adenine dinucleotide in reduced form(NADH)was determined to obtain the velocity of the enzymatic reaction and the calibration curve of ethanol was drawn.The ethanol in samples was determined by the calibration curve.The effect of the pH and the inhibitor was discussed.The method is simple,rapid and accurate.
