摘要
目的 探讨蓝舌病毒湖北株 (BTV -HbC)对人癌细胞的杀伤作用。方法 采用病毒学、细胞培养、透射电镜和免疫学技术观察BTV -HbC对人正常二倍体细胞和人癌细胞的感染。结果 BTV -HbC对正常二倍体人胚肺HEL细胞不感染 ,却能在实验组肿瘤细胞中选择性增殖 ,产生不同程度细胞病变效应 (CPE ) ,终致肿瘤细胞死亡。其中 ,人肺癌SPC -A1细胞最敏感[CPE出现时间早 ,病毒接种后 3 6h细胞死亡数达 90 % ,TCID50 为 10 -3 ,透射电镜下该肿瘤细胞质内BTV -HbC颗粒量最多 ,并可见包涵体 (inclusionbody)形成 ] ,人宫颈癌HeLa细胞次之 ,人星形胶质瘤U2 51细胞再次之 ,而小鼠星形胶质瘤C6细胞敏感性最差。琼脂糖双向扩散试验进一步证实了肿瘤细胞内皆有BTV -HbC存在。同时 ,透射电镜下实验组 4株肿瘤细胞内皆可见核膜下染色质边缘化 ,高度凝集 ,细胞固缩等类似细胞凋亡 (apoptosis)现象。 结论 不感染人正常二倍体细胞的BTV -HbC ,却可以在人肿瘤细胞中选择性增殖产生CPE ,并诱导凋亡 。
Objective To explore bluetongue virus HbC strain(BTV-HbC) infection of human cancer cell lines.Methods Using the virological technique,cell culture,transmission election microscopy(TEM) and immunological technique,we observed bluetongue virus HbC strain(BTV-HbC) infection of human cancer cell lines.Results Under identical conditions,BTV-HbC couldn′t infect HEL cells,while they could replicate selectively in four types of experimental tumor cells,producing different cytopathic effect(CPE) and eventually caused complete lysis and death of tumor cells.SPC-A 1 cells were the most sensitive type(CPE occurred earliest and 90 percent of cells were killed after being inoculated with BTV-HbC strain for 36 hours,TCID50 was 10 -3 and the BTV-HbC virions existing in the cytoplasm of tumor cells were the largest under TEM.HeLa was the second one,U 251 the third and C 6 the least.Agarose two-direction diffusing test proved the existing of BTV-HbC in the tumor cells.In addition,under TEM,it was found that some apoptosis were occurred in each type of tumor cells.Conclusion BTV-HbC strain could not infect human normal diploid cell but could selectively replicate in the tumor cells,producing CPE,inducing apoptosis and eventually causing the death of tumor cells.
