摘要
目的 研究奥曲肽对胃癌细胞系SGC -790 1生长的抑制作用。方法 将奥曲肽注入胃癌细胞 ,采用四甲基偶氮唑蓝 (MTT )法测定细胞生长抑制效应 ,将3 H -胸腺嘧啶核苷 (3 H -TdR )掺入试验测定奥曲肽对胃癌细胞DNA合成的影响 ,显微镜下观察胃癌细胞形态变化 ,应用流式细胞仪 (FCM )检测细胞周期 ,并采用TRAPPCR -ELISA法对胃癌细胞端粒酶活性进行检测。结果 奥曲肽对胃癌细胞系SGC -790 1的生长及DNA合成均有抑制作用 ,显微镜下可见细胞变圆、变小、脱落 ,FCM检测结果显示奥曲肽作用后 ,G0 /G1期细胞比例增高 ,S期、G2 /M期细胞比例下降 ,端粒酶活性显著受抑制。结论 奥曲肽能明显抑制胃癌细胞系SGC -790 1的生长 ,形成G0 /G1期阻滞 。
Objective To study inhibition effect of octreotide on the growth of gastric carcinoma cells SGC-7901.Methods The gastric carcinoma cells were treated with octreotide.The effect of growth suppression on cells was evaluated by MTT assay.The effect of octreotide on DNA synthesis of gastric carcinoma cells was evaluated by 3H-thymidine( 3H-TdR)incorporation.Morphologic changes of gastric carcinoma cells were observed under microscope.Cell cycle was detected by flow cytometry(FCM).The activity of telomerase of gastric carcinoma cells was examined by TRAP PCR-ELISA.Results Octreotide obviously inhibited the growth and DNA synthesis of SGC-7901 cells.SGC-7901 cells became round,small,and exfoliating after treated with Octreotide.The FCM analysis showed that the number of gastric carcinoma cell of G 0/G 1 phase increased and the number of cell of S phase and G 2/M phase decreased.The activity of telomerase of gastric carcinoma cells was obviously inhibited.Conclusion Octreotide can obviously inhibit the growth of SGC-7901 cells,cause G 0/G 1 blocking and inhibit the activity of telomerase.
