高效液相色谱法测定复方达美肤溶液及皮炎灵中氯倍他索的含量

Determination of clobetasol in Fufangdameifu solution and Piyanling by HPLC

目的 :建立复方达美肤溶液和皮炎灵中氯倍他索的含量测定方法。方法 :采用高压液相色谱法 (HPLC) ,NUCLEOSIL 10 0 5C18柱 ( 3.9mm×15 0mm ,5 μm) ,以甲醇∶0 .0 5mol·L- 1醋酸/醋酸钠缓冲液 =78∶2 2 (v∶v)为流动相 ,流速 1.0mL·min- 1,检测波长 2 37nm。结果 :复方达美肤溶液中氯倍他索在 0 .4 0～ 0 .60 g·L- 1范围内呈良好的线性关系 (r =0 .999,n =5 ) ;皮炎灵中氯倍他索在0 .12～ 0 .18g·L- 1范围内呈良好的线性关系 (r =0 .999,n =5 )。结论 :HPLC测定复方达美肤溶液、皮炎灵中氯倍他索的方法准确、快速、可行。

AIM:To establish HPLC method for the determination of clobetasol in Fufangdameifu and Piyanling. METHODS:The clobetasol was assayed by HPLC with a NUCLEOSIL 100-5C 18 column(3.9 mm×150 mm,5 μm). The mobile phase was consisted of methanol∶0.05 mol·L -1 HAC/NaAC buffer(78∶22).The flow rate was 1.0 mL·min -1. The detection wavelength was 237 nm. RESULTS:The linear range of clobetasol in Fufangdameifu was 0.4～ 0.6 g·L -1(r=0.999,n=5), and that in Piyanling was 0.12～0.18 g·L -1(r=0.999,n=5). CONCLUSION: The HPLC method for the determination of clobetasol in Fufangdameifu and Piyanling is accurate, rapid, and practicable.
