摘要
目的通过检测脑胶质瘤相关基因1(Gli 1)与下调叉头框蛋白M1(FoxM 1)在正常宫颈组织、子宫颈上皮不典型增生(CIN)组织、宫颈癌中表达情况及其相关性,研究探讨两者在宫颈癌发生中的作用及临床意义。方法通过免疫组织化学方法检测正常宫颈组织、CIN组织及宫颈癌标本中的Gli 1与FoxM 1的蛋白含量,比较三种标本中的Gli 1与FoxM 1的蛋白表达阳性率差异及两者相关性分析,进而探讨两种蛋白对宫颈疾病的影响。结果正常宫颈组织、CIN组织、宫颈癌标本中Gli 1蛋白表达量分别为:5%、35%、69%,CIN组织及宫颈癌患者的Gli 1蛋白表达均高于对照组,差异均有统计学意义(χ~2=26.72,73.74,P<0.05);宫颈癌患者Gli 1蛋白表达高于CIN组织患者,差异有统计学意义(χ~2=15.52,P<0.05);正常宫颈组织、CIN组织、宫颈癌标本中FoxM 1蛋白表达量分别为:6%、55%、88%,CIN组织及宫颈癌患者的FoxM 1蛋白表达均高于对照组,差异均有统计学意义(χ~2=53.19、106.85,P均<0.05);宫颈癌患者FoxM 1蛋白表达高于CIN组织患者,差异有统计学意义(χ~2=17.00,P<0.05);Gli 1与FoxM 1两种蛋白在宫颈癌组织中的表达相关(r=0.897,P<0.05)。结论 FoxM 1和Gli 1与宫颈癌发生发展相关,可为宫颈癌的早期诊断及治疗提供理论依据。
Objective To detect the expression of Glioma-associated oncogene 1(Gli 1)and Forkhead box M1(FoxM 1)in different types of cervical tissues,and evaluate its clinical significance.Methods The protein content of Gli 1 and FoxM 1 in normal cervical tissue,cervical intraepithelial neoplasia(CIN)tissue and cervical cancer specimens was detected by immunohistochemistry.The differences in the positive rates of protein expression between Gli 1 and FoxM 1 in the three specimens and their correlation were analyzed.The effects of two proteins on cervical diseases were also explored.Results Gli 1 protein expression rates in normal cervical tissue,CIN tissue and cervical cancer specimens were 5%,35%and69%,respectively.The Gli 1 protein expression levels of CIN tissue and cervical cancer were both higher than that of compaired group;this difference had a clinical significance(χ~2=26.72,73.74,P<0.05).The expression level of Gli 1 protein in cervical disease was higher than that in CIN tissue;this difference had a clinical significance(χ~2=15.52,P<0.05).FoxM 1 protein expression rates in normal cervical tissue,CIN tissue and cervical cancer specimens were 6%,55%and 88%,respectively.The FoxM 1 protein expression levels of CIN tissue and cervical cancer were both significantly higher than that of compaired group(χ~2=53.19,106.85,P<0.05).The expression level of FoxM 1 protein in cervical disease was significantly higher than that in CIN tissue(χ~2=17.00,P<0.05).The expressions of Gli 1 and FoxM 1 protein in cervical cancer tissues were correlative(r=0.897,P<0.05).Conclusion FoxM 1 and Gli 1 were related to the development of cervical cancer,which could provide a theoretical basis for the early diagnosis and treatment of cervical cancer.
