慢性萎缩性胃炎患者胃黏膜组织中RKIP,STAT3和CyclinD1的表达变化及意义 被引量:9

The expression changes and significances of RKIP, STAT3 and CyclinD1 in gastric mucosal tissues of chronic atrophic gastritis

摘要 目的探讨慢性萎缩性胃炎(CAG)胃黏膜组织中Raf-1激酶抑制蛋白(RKIP),信号转导子和转录激活子3(STAT3)和细胞周期素D1(CyclinD1)的表达变化及意义。方法选取2017年9月至2018年8月于本院进行胃镜检查的86例患者为研究对象,分为CAG组53例,慢性浅表性胃炎(GSG)组14例,正常胃黏膜组(对照组)19例。采用实时荧光定量PCR(q RT-PCR)检测各组RKIP、STAT3、CyclinD1 mRNA表达水平;免疫组化法检测RKIP、STAT3、CyclinD1蛋白表达。Pearson法分析CAG组RKIP、STAT3、CyclinD1表达的相关性。结果与GSG组、对照组相比,CAG组RKIP m RNA表达水平及蛋白阳性表达率均显著降低,且GSG组显著低于对照组,差异有统计学意义(P<0.05);CAG组STAT3、CyclinD1 mRNA表达水平及蛋白阳性表达率均显著高于GSG组、对照组,且GSG组显著高于对照组,差异有统计学意义(P<0.05);Pearson分析显示CAG组RKIP表达均与STAT3、CyclinD1表达呈显著负相关,差异有统计学意义(P<0.05),STAT3与CyclinD1表达呈显著正相关,差异有统计学意义(P<0.05)。结论慢性萎缩性胃炎患者胃黏膜组织中RKIP表达水平降低,而STAT3与CyclinD1表达水平升高,三者均可能参与CAG发生及发展过程,并可作为临床早期诊断CAG的重要辅助指标。 Objective To investigate the expression changes and significances of Raf-1 kinase inhibitor protein(RKIP),signal transducer and activator of transcription 3(STAT3)and cyclinD1(CyclinD1)in gastric mucosal tissues of chronicatrophic gastritis(CAG).Methods 86 patients who underwent gastroscopy in our hospital from September 2017 to August2018 were selected as the subjects,and divided into CAG group(53 cases),chronic superficial gastritis(GSG)group 14 cases and normal gastric mucosa group(control group)19 cases.The expression levels of RKIP,STAT3 and CyclinD1 mRNAs in each group were detected by real-time fluorescence quantitative PCR(qRT-PCR);the expressions of RKIP,STAT3 and CyclinD1 were detected by immunohistochemistry.Pearson method was used to analyze the correlations betweenthe expressions of RKIP,STAT3 and CyclinD1 in CAG group.Results Compared with GSG group and control group,theexpression level of RKIP mRNA and protein positive rate in CAG group were significantly decreased(P<0.05),and GSGgroup was significantly lower than control group(P<0.05);the expression levels of STAT3 and CyclinD1 mRNA and proteinpositive rates in CAG group were significantly higher than those in GSG group and control group(P<0.05),and GSG groupwas significantly higher than control group(P<0.05);Pearson analysis showed that the expression of RKIP in CAG groupwas negatively correlated with the expressions of STAT3 and CyclinD1(P<0.05),and STAT3 was positively correlated withthe expression of CyclinD1(P<0.05).Conclusion The expression level of RKIP in gastric mucosa was decreased inpatients with chronic atrophic gastritis;the expression level of STAT3 and CyclinD1 were increased.All of them might beinvolved in the occurrence and development of CAG,and could be used as an important auxiliary index for early clinicaldiagnosis of CAG.
