丝裂霉素、5-氟尿嘧啶、三苯氧胺联合应用对人乳癌细胞株的作用

摘要 目的:观察丝裂霉素、5-氟尿嘧啶、三苯氧胺联合应用对人乳腺癌细胞株(MDA-MB-435S)的相互作用。方法:采用MTT法利用中效原理判断联合用药的效应。结果:MMC、5-Fu单用及联合应用时,随着药物浓度增加其效应也增加;TAM与MMC、5-Fu合用时,对MDA-MB-435S细胞作用不明显;MMC与5-Fu高浓度合用时呈协同作用,低浓度时呈拮抗作用;两药合用时浓度比例变化及给药时间次序对效应影响不大。结论:MMC与5-Fu高浓度合用时为协同作用,低浓度时为拮抗作用。TAM与MMC、5-Fu合用时,对MDA-MB-435S细胞作用不明显。两药合用效应大小与两药浓度比例及给药时间次序无关。 Objective: To evaluate the killing effects of MMC, 5 - Fu and TAM on human breast cancer cell line(and MDA-MB-435S) .Methods: The median - effect principle was used. Results: Killing effects of the individual drugs were enhanced as median concentration increased. We also observed that the interaction of drugs was synergy at higher concentration (CI< 1) . Finally, both the ratios of drug concentration and the sequence of administration were not related with the killing effects. Concluion: The interaction of drugs was synergy at higher concentration.The drug administration order and drug concentrations are not related with the killing effects.
