摘要
目的:建立羊开口药材HPLC特征图谱的质量评价方法,并比较不同基源的羊开口HPLC特征图谱。方法:色谱柱为Phenomenex C18(250.0 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液,体积流量1.0 mL/min,检测波长364 nm,柱温35℃,进样量20μL。结果:从羊开口药材特征图谱中分析得出6个共有峰,其中2号峰为鞣花酸,4号峰为3,3',4'-三甲氧基鞣花酸-4-O-β-D-葡萄糖苷。结论:本研究建立的羊开口药材的特征图谱特征性和专属性强,且展毛野牡丹和野牡丹的特征图谱相似度较高,这为将野牡丹和展毛野牡丹作为羊开口药用基源开发提供了科学依据。
Objective:To establish a quality evaluation method for the characteristic chromatograms of Melastoma Radix Et Rhizoma by HPLC,and to compare the characteristic chromatograms from different sources.Methods:The column was Phenomenex C18(250.0 mm×4.6 mm,5μm)with the mobile phase of acetonitrile-0.1%phosphoric acid at the flow rate of1.0 m L/min;the detection wavelength was 364 nm;the column temperature was 35℃and the sample size was 20μL.Results:There were six common peaks identified in the characteristic chromatograms of Melastoma Radix Et Rhizoma,among which Peak 2 was ellagic acid and Peak 4 was 3,3’,4’-tri-omethlellagic acid-4-O-β-D-glucoside.Conclusion:The characteristic chromatograms of Melastoma Radix Et Rhizoma established in this study are obviously characteristic and specific,and the characteristic chromatograms of Melastoma Normale D.Don and Melastoma Candidum D.Don show high similarity,which provides a scientific basis for the development of Melastoma Candidum D.Don and Melastoma Normale D.Don as the sources of Melastoma Radix Et Caulis.
