右归饮联合盐酸雷洛昔芬对骨质疏松症小鼠骨形成、骨密度影响的研究 被引量:1

Yougui Yin Combined with Raloxifene Hydrochloride Has Effect on Bone Formation and Bone Density in Mice with Osteoporosis

摘要 目的:探究右归饮联合盐酸雷洛昔芬对骨质疏松症小鼠骨形成、骨密度的影响。方法:选取40只C57BL/6品系雌性小鼠,切除卵巢构建骨质疏松症模型,饲养12周,分为假手术组、模型组、雷洛昔芬组及联合组。其中假手术组和模型组均用0.5 mL0.9%氯化钠溶液灌胃,雷洛昔芬组用5 mg/kg盐酸雷洛昔芬治疗,联合组用0.2 m L右归饮+5 mg/kg盐酸雷洛昔芬治疗。连续给药6周后眼球取血,检测血清骨钙素(Osteocalcin,OC)、I型胶原氨基端延长肽(Procollagen I N-terminal Propeptide,PINP)、碱性磷酸酶(Alkaline phosphatase,ALP)、抗酒石酸酸性磷酸酶(Tartrate-resistant acid phosphatase,TRACP)等骨代谢指标水平。处死小鼠,测定骨密度,Micro-CT检测骨体积分数(Bone volume/total volume,BV/TV)、骨表面积/骨体积(Bone surface/bone volum,BS/BV)、骨小梁分离度(Trabecular separation/spacing,Tb.Sp)、骨小梁平均厚度(Trabecular thickness,Tb.Th)等骨组织形态指标。胫骨组织HE染色,观察骨组织形态损伤情况。结果:与假手术组比较,模型组小鼠骨密度、BV/TV、Tb.Th、PINP降低,差异均有统计学意义(P <0.05),BS/BV、Tb.Sp、OC、ALP、TRACP升高,差异均有统计学意义(P <0.05);与模型组比较,雷洛昔芬组、联合组骨密度、BV/TV、Tb.Th、PINP升高,差异均有统计学意义(P <0.05),BS/BV、Tb.Sp、OC、ALP、TRACP降低,差异均有统计学意义(P <0.05);与雷洛昔芬组比较,联合组骨密度、BV/TV、Tb.Th、PINP升高,差异均有统计学意义(P <0.05),BS/BV、Tb.Sp、OC、ALP、TRACP降低,差异均有统计学意义(P <0.05)。结论:右归饮联合盐酸雷洛昔芬治疗骨质疏松症比单用盐酸雷洛昔芬能更显著地抑制骨丢失,改善骨代谢,促进骨形成,达到增加骨密度的治疗效果。 Objective:To explore the effect of Yougui Yin combined with raloxifene hydrochloride on bone formation and bone density in mice with osteoporosis.Methods:Forty female C57 BL/6 strain mice whose ovaries were removed to establish models with osteoporosis.The mice were reared for twelve weeks and divided into the sham group,the model group,the raloxifene group and the combination group.The sham group and the model group were treated with 0.5 ml 0.9%sodium chloride solution;the raloxifene group was treated with 5 mg/kg raloxifene hydrochloride,and the combination group was treated with 0.2 ml Yougui Yin plus 5 mg/kg raloxifene hydrochloride.After six weeks of treatment,blood of eyeballs was taken to detect Osteocalcin(OC),Procollagen I N-terminal Propeptide(PINP),Alkaline phosphatase(ALP)and Tartrate-resistant acid phosphatase(TRACP).After that,the mice was put to death and then the bone density was observed,and morphological indexes of bone tissue including Bone volume/total volume(BV/TV),Bone surface/bone volum(BS/BV),Trabecular separation/spacing(Tb.Sp)and Trabecular thickness(Tb.Th)were detected through Micro-CT.HE staining of tibial tissue was carried out to observe the morphological damage of bone tissue.Results:Compared with those in the sham group,the bone density,BV/TV,Tb.Th and PINP were decreased,differences being significant(P<0.05),while BS/BV,Tb.Sp,OC,ALP and TRACP were increased,differences being significant(P<0.05).Compared with those in the model group,the bone density,BV/TV,Tb.Th and PINP in the raloxifene group and the combination group were increased,differences being significant(P<0.05),while BS/BV,Tb.Sp,OC,ALP and TRACP were decreased,differences being significant(P<0.05).Compared with those in the raloxifene group,the bone density,BV/TV,Tb.Th and PINP in the combination group were increased,differences being significant(P<0.05),while BS/BV,Tb.Sp,OC,ALP and TRACP were decreased,differences being significant(P<0.05).Conclusion:Yougui Yin combined with raloxifene hydrochloride can more significantly inhibit bone loss,improve bone metabolism,promote bone formation and increase bone density in the treatment of osteoporosis than raloxifene hydrochloride alone.
