目的:建立一种反相高效液相色谱一荧光检测法测定血清中异丙酚浓度。方法:用美国Spectra—Physics高效液相色谱系统,色谱柱为Diamonsil C_(18)柱,以乙腈-水(70:30,含0.1%三氟乙酸)为流动相,荧光激发波长276 nm,发射波氏310 nm。饱和硫酸铵作蛋白沉淀剂,血清中加入内标麝香草酚,含有内标的乙腈溶液作为异丙酚提取剂。结果:本方法测定的线性范围0%25~10 mg·L^(-1),最低检出浓度25 μg·L^(-1),加标回收率为91.2%~100.9%,日内精密度≤1.50%,日间精密度≤2.51%。结论:本方法较为简单,结果准确可靠,适用于异丙酚的药代动力学研究及临床手术中异丙酚药物浓度的监测。
OBJECTIVE: To develop a RP HPLC method for determining serum concentration of propofol. METHODS: The propofol was separated on a Diamonsil C_(18) column, the mobile phase was a mixture of acetonitrile and water (70:30, containing 0.10% trifluoroacetic acid). The excitation and emission wavelength were 276nm and 310nm, respectively. Sample preparation was simply a deproteinsation process with saturated ammonium sulphate. The propofol was extracted by the acetonitrile, the solvent of internal standard (thymol). RESULTS: A good linear relationship was obtained between 0.25 - 10 mg · L^(-1) (γ=0.9996). The detection limit was 25 Mg·L^(-1) with an injection volume of 20 μ L. The RSD of within-day and between-day were less than 2.5%, the average recovery was 91.2% - 100.9%. CONCLUSION: The proposed method is sensitive, reliable and can be used for pharmacokinetic studies of propofol and monitoring propofol in serum.
