低硒心肌损伤与硫氧还蛋白还原酶的关系研究 被引量:3

摘要 目的 研究长期低硒心肌硫氧还蛋白还原酶的活性和表达改变 ,探讨硫氧还蛋白还原酶与心肌损伤的关系。方法 用低硒饲料喂养Wistar大鼠 14周 ,通过透射电镜观察心肌的病理学改变 ,采用生物化学方法和Westernblot杂交检测心肌组织中硫氧还蛋白还原酶的活性及蛋白表达。结果 低硒导致大鼠心肌线粒体轻度损伤。低硒组大鼠心肌硫氧还蛋白还原酶活性降低至对照组的 6 3.8% ,经t检验P <0 .0 1。蛋白杂交扫描结果显示 ,常硒组峰面积为 3438,低硒组峰面积为 2 0 6 7。结论 低硒降低硫氧还蛋白还原酶的活性和蛋白表达水平 ,使心肌抗氧化能力减低 ,引起心肌损伤。 Objective To study the changes of activity and expression of thioredoxin reductase in the selenium deficient rats.Analyse the relationship between thioredoxin reductase and cardiomyocytes injury.Methods Wistar rats were fed selenium deficient diets for 14 weeks.Pathological changes were observed by transmission electronic microscope.The changes of thioredoxin reductase activity and protein expression were detected by biochemistry and western blotting.Results Selenium deficiency induced slight myocardium mitochondria injury in rats.Selenium deficiency caused a decrease in activity from control to 63.8% in cardiomyocytes.The result of t test was P <0.01.By the results of protein hybridization scan, peak area in selenium adequate group was 3 438 and in selenium deficient group was 2 067.Conclusion Selenium deficiency decreases the activity and protein levels of thioredoxin reductase in cardiomyocytes,and decreases cardiomyocytes anti oxidation and induce cardiomyocytes injury.
