摘要 目的 研究荧光金在视神经损伤中的应用。方法 荧光金注射到正常及不完全视神经损伤后大鼠的外侧膝状体和上丘 ,观察视网膜铺片中视网膜神经节细胞的形态和数量 ;荧光金注射到大鼠玻璃体腔内 ,观察视网膜神经节细胞轴突的损伤情况。结果 视网膜铺片的节细胞边界清晰 ,易于计数和形态的观察。损伤 2周时视网膜神经节细胞剩余 5 7% ,相对应的损伤 2周的轴突远端的线形荧光明显减少。结论 荧光金是视神经损伤中视网膜神经节细胞数目变化和轴突损伤、分布和投射的可靠有效方法。 Objective To explore the use of fluorogold in the research on crush wound of optic nerves. Methods Firstly, rat model was induced by means of incompletely crush wound of optic nerve using across action forceps at 2mm behind eyeball.Then we oberserved the injury of axon by both anterograde and retrograde tracing of fluorogold.Results RGCSs of retina was easy to observe and count. 57% of RGCSs remained alive and the distant axon decreased at 2 weeks. Conclusion Anterograde and retrograde tracing of fluorogold do help in the research of wound of optic nerves.
