摘要
目的 探讨肝细胞癌变过程中cyclinD1 的异常表达及其意义。方法 用 0 .0 5 %的 2 AAF诱导Wistar大鼠建立肝癌动物模型 ,应用免疫组化和原位杂交法检测大鼠肝脏在癌变过程中cyclinD1 的蛋白和mRNA表达。结果 正常组 8只中cyclinD1 蛋白和mRNA表达均正常 ,肝硬化组 16只中有 12只cyclinD1 蛋白和mRNA过表达 ,肝癌组 11例中有 4只cyclinD1 蛋白和mRNA过表达。结论 在 3组中cyclinD1 蛋白和mRNA表达一致 ,无显著差异。肝硬化组和肝癌组的cyclinD1 异常表达均高于正常组 ,而且肝硬化组的cyclinD1 异常表达高于肝癌组。
Objective To study the expression and significance of cyclin D 1 during hepatocellular canceration. Methods 0.05% 2 AAF induced rat(Wister) hepatocellular carcinoma. Cyclin D 1 protein and mRNA were examined respectively by immunohistochemistry and in situ hybridization techniques,respectively. Results All of control group showed normal expression of cyclin D 1 protein and mRNA, 12 of 16 liver cirrhosis cases showed overexpression of cyclin D 1 protein and mRNA.Four of 11 hepatocellular carcinoma cases showed overexpression of cyclin D 1 protein and mRNA in the three groups. Conclusion There is no significant difference between expressions of cyclin D 1 protein and mRNA. Cyclin D 1 overexpression in liver cirrhosis and hepatocellular carcinoma shows higher than those in control group. Cyclin D 1 overexpression in liver cirrhosis shows higher than those in hepatocellular carcinoma.
