摘要
酶解法提油具有提取率高、反应条件稳定、有效成分损失少等优点,本文通过对酶解法提油工艺进行改进,采用水酶法对斑点叉尾鮰鱼内脏油进行提取并对提取工艺参数进行响应面优化。在液料比、加酶量、酶解PH、温度、时间5个单因素试验的基础上,通过数学模型优化的最佳提取工艺参数为:时间75.2min、液料比(mL/g) 1∶1、温度61.51℃、pH7.23、加酶量3 000.06U/g,预测鱼油提取率为87.0%;为便于操作,确定最佳的提取工艺参数为:时间75min、液料比(mL/g) 1∶1、温度61.5℃、pH7.2、加酶量3 000U/g。在最佳条件下,得到的鱼油提取率为86.34%,实际值与预测值基本一致,这表明了响应面法优化得到的最佳工艺参数可信度较高。
Enzymatic extraction of oil had the advantages including high extraction rate,stable reaction conditions and less loss of effective components.The technology of enzymatic extraction of oil was improved,and the aqueous enzymatic extraction was adopted,and the viscera oil of ietalurus punetaus was extracted,and the extraction technology parameters were optimized by response surface.On the basis of five single factor experiment including liquid-solid ratio,enzyme dosage,enzymatic hydrolysis pH,temperature and time,the optimum extraction technology parameters optimized by mathematical model were as follows:the time was 75.2 min,the liquid-solid ratio was mL/g 1∶1,and the temperature was 61.51°C,and the pH was 7.23,and the enzyme dosage was 3 000.06 U/g.The fish oil extraction rate was predicted to be 87.0%.In order to facilitate the operation,the optimum extraction technology parameters were determined and it was as follows:the time was 75 min,and the liquid-solid ratio was mL/g 1∶1,and the temperature was 61.5°C,and the pH was 7.2,and the enzyme dosage was 3 000 U/g.Under the optimum conditions,the fish oil extraction rate was 86.34%,and the actual value was basically consistent with the predicted value,which indicated that the optimum technology parameters obtained by response surface method were highly reliable.
