对小鼠糖尿病模型胰岛素治疗的研究

Study on Insulin Treatment for Mouse Diabetes Model

目的 探讨适宜的小鼠糖尿病模型的制作方法及对小鼠糖尿病模型的用药治疗。方法 利用链脲菌素制作小鼠模型 ,找出适宜的注射浓度及注射量 ,将制作好的小鼠糖尿病模型给药治疗 ,研究用药的最佳治疗方案。结果 利用 10g L的链脲菌素溶液对小鼠腹腔注射 ,注射量为 0 3ml的小鼠血糖上升不明显 ,短期内无法制作成模型。注射量为 0 5ml小鼠死亡率高 ,不适宜。注射量为 0 4ml的小鼠短期内血糖升至 2 0mmol L以上 ,为成功糖尿病模型。糖尿病小鼠给药治疗 ,设计了 5种治疗方案 ,以第 5个方案为佳。结论 注射量为 0 3ml因量小 ,小鼠血糖上升不明显 ;注射量为 0 5ml的小鼠因注射量过大 ,水中毒死亡 ,因此 ,0 4ml为最佳注射量。治疗方案 5能够给糖尿病小鼠提供一个较平稳的基础胰岛素浓度 ,治疗结果较满意。

Objective Probe the proper way of making the model of mouse diabetes and the medical treatment for mouse diabetes model.Methods Make the mouse model with streptozotocin to find proper concentration and dose of injection.Give medical treatment for the mouse diabetes model which has already made,and study the best plan of medical treatment.Result Inject mouse abdominal cavity with 10g/L streptozotocin solution.The blood sugar of mouse,whose injection dose was 0 3ml didn't rise obviously.We can't make successful model with them in a short period of time.The mortality rate of those whose injection dose was 0 5ml was high.They were not proper to make model with them.The blood sugar of those whose injection close was 0 4ml rose higher than 20mmol/L,they were successful mouse diabetes model.Five plans of medical treatment for diabetes mice were made,the fifth one was the best one.Conclusion The quantity of whose injection dose was 0 3ml was small,the blood sugar of these mice didn′t rise obviously .The mice whose injection dose was 0 5ml died because of poisoning,owing to excessive injection dose.Therefore,0 4ml injection dose is the best one.The fifth plan of treatment can provide a smoother basic insulin concentration for diabetes mice.The result of treatment is more satisfied.
