摘要
目的 :观察氯化钴 (CoCl2 )预处理对急性低氧后海马神经元电压门控性Na+ 、K+ 电流的影响。方法 :原代培养大鼠海马神经元 ,分为CoCl2 预处理和非处理组 ,采用膜片钳全细胞记录技术 ,检测急性低氧后海马神经元钠电流(INa)、钾电流 (Ik)的变化。结果 :急性低氧后 ,海马神经元INa、IK 电流幅度明显降低 ,INa阈值右移 ,而经CoCl2 预处理的海马神经元INa、IK 电流的降低幅度明显减轻。结论 :CoCl2 预处理减轻急性低氧所致的INa、IK 电流变化 。
Aim: To study effect of CoCl 2 pretreatment on the voltage gated Na + and K + currents of the rat hippocampal neurons after acute hypoxia. Methods: Primarily cultured hippocampal neurons were divided into CoCl 2 pretreated and non pretreated groups. Patch clamp whole cell recording technique was used to examine Na + and K + currents of the hippocampal neurons. Results: After acute hypoxia, I Na and I K of the hippocampal neurons were significantly decreased and the threshold of I Na was right shifted. Pretreatment of the neurons with CoCl 2 inhibited the reduction of I Na and I K. Conclusion: CoCl 2 pretreatment alleviates the acute hypoxia induced changes of I Na and I K, which may be one of the mechanisms for the protective effect of CoCl 2 on neurons.
