摘要
目的:研究小鼠大脑中动脉闭塞(MCAO)后不同运动训练方式对大脑皮质胆碱能(Ach)纤维密度的影响。方法:45只雄性C57BL/6J小鼠,建立小鼠MCAO模型。将MCAO小鼠随机分为运动1组、运动2组、对照组。运动1组跑笼训练1小时/天,共90天,运动Ⅱ组跑笼训练15天,停止30天,再跑笼45天;对照组跑笼固定不转。分别在MCAO后15、45、90天;观察小鼠大脑皮质Ach纤维密度改变。结果:MCAO后运动15天后,各组皮质Ⅲ、Ⅴ层Ach纤维密度无显著差异(P>0.05)。运动45天后,运动1组比对照组在皮质Ⅲ、Ⅴ层正常侧Ach纤维密度显著增高(P<0.05),比运动2组皮质Ⅲ层正常侧Ach纤维密度高(P<0.05),在缺血侧,3组间无明显差异(P>0.05)。90天后,运动1组与运动2组及对照组比较,在正常侧皮质Ⅲ、Ⅴ层纤维密度均增高(P<0.05),运动1组比对照组缺血侧皮质Ⅲ、Ⅴ层纤维密度均高(P<0.05),运动1组比运动2组皮质Ⅴ层纤维密度高(P<0.05),运动2组比对照组缺血侧皮质Ⅴ层纤维密度增高(P<0.05)。结论:运动训练促进了小鼠MCAO后双侧大脑皮层胆碱能纤维的发芽,恢复期训练仍有一定作用。
Objective:To investigate the impact of different form of physical training on the AchE-positive fibers density after middle cerebral artery occlusion(MCAO)in mice.Method:Forty-five male C57BL/6J mice underwent MCAO and then were divided into training group1(n=15),training group2(n=15)and control group(n=15).The mice of training group1were trained with voluntary running wheel for one hour daily for90days.The mice of training group2were trained for15days,stopped for30days and retrained for45days after MCAO.The running wheel of control group was sedentary.Fifteen days,45days and90days after MCAO,AchE-positive fibers density were studied respectively in cerebral cortex.Result:There was no difference about AchE positive fibers inⅢ,Ⅴlayers of cortex in all groups in15days after MCAO(P>0.05).In45days after MCAO,AchE-positive fibers density of training group1showed a significant increase inⅢ,Ⅴlayers of cortex in normal side than it in control group(P<0.05)and only inⅢlayers of cortex than it in training group2(P<0.05).No significant density difference in found in lesion side in all groups(P>0.05).In90days after MCAO,compared with training group2and control group AchE-positive fibers density of training group1increased inⅢ,Ⅴlayers of cortex in normal side(P<0.05).In lesion side,compared with control group AchE-positive fibers density of training group1increased inⅢ,Ⅴlayers(P<0.05)and AchE-positive fibers density of training group2only increased inⅤlayers(P<0.05).Conclusion:The physical training can modify the loss of bilateral cortical cholinergic fibers in mice after MCAO.Training in recovery period after MCAO can also increase the AchE-positive fibers density to a certain extent.
