摘要
采用福林法测定了藕节中总酚的含量 ,确定了以没食子酸为标准品福林法测定藕节中总酚的适宜方法。结果表明 ,没食子酸在福林试剂与 10 %Na2 CO3 之比为 1∶1(v∶v) ,体积为 3ml,反应温度为 2 5℃ ,反应时间为 30min ,浓度为 2~ 10 μg/ml时与其吸光值呈良好的线性关系。测定方法的回收率为 89.5 %~ 96 .5 % ,变异系数为 2 .14 % ,有较好的精确度和准确度。
In this research, the optimum conditions for determining of the total phenol in the joint of lotus roots were investigated. The results showed that when the reaction conditons were volume fraction 1∶1(v∶v),volume 3 ml,temperature 25 ℃,operation time 30 min,there was linear relationship between gollic acid and absorbance value over the range of 2~10 μg/ml,the rate of recovery of the total phenolics content ranges from 89.5% to 96.5%,the variation coefficient is 2.14%.It is a kind of correct and accurate method to determinate the total phenol content.
