摘要
以斑马鱼为模型,研究番红花花提取物对紫外线诱导斑马鱼鱼鳍损伤的保护作用。将受精后72 h的斑马鱼幼鱼经番红花花提取物溶液处理,同时进行UVB照射后,观察斑马鱼鱼鳍的损伤程度,以及通过吖啶橙染色检测斑马鱼鱼鳍的凋亡细胞。由实验结果可知,在UVB照射后的第1天和第2天,与未照射阴性对照组(-UVB)相比,模型对照组(+UVB)的斑马鱼鱼鳍都出现明显的损伤,而200 mg/L番红花花提取物处理组(+UVB)的斑马鱼鱼鳍有较少的损伤,而且400 mg/L和800mg/L番红花花提取物处理组(+UVB)的斑马鱼鱼鳍几乎没有损伤,与阴性对照组相似;吖啶橙染色结果显示,模型对照组(+UVB)的斑马鱼鱼鳍有大量的凋亡细胞,而实验组(番红花花提取物+UVB)的斑马鱼鱼鳍仅有少量的凋亡细胞,与阴性对照组(-UVB)相似;番红花花提取物对紫外线诱导斑马鱼鱼鳍损伤具有保护作用。
The protective effect of crocus sativus flower extract against UV was evaluated in a zebrafish model by recording fin morphological changes.The damage degree of zebrafish fins was observed for the zebrafish at 72 h after treated with crocus sativus flower extract and UVB irradiation.The cell death of zebrafish fins was detected by acridine orange fluorescent staining.On days 1 and 2 after UVB irradiation,the zebrafish fins in UVB-only group showed significant damage compared with the non-irradiated negative control group,while the fins in the UV+200 mg/L crocus sativus flower extract experimental group had less damage.Moreover,the fins in the UV+400 mg/L and 800 mg/L crocus sativus flower extract experimental groups almost had no damage,which was similar to the negative control group.In addition,the results of acridine orange fluorescent staining showed that the UVB-only group had a large number of apoptotic cells in the zebrafish fin,while the UVB+crocus sativus flower extract experimental group had only a small amount of apoptotic cells in the zebrafish fins,which was similar to the negative control group.The crocus sativus flower extract had protective effect on the zebrafish fins from UV-induced damage.
