摘要
在pH为 1 8的Clark Lubs缓冲溶液条件下 ,人血清白蛋白与靛红结合 ,使靛红的吸光度下降 .质量浓度在 6~ 34mg/L范围内 ,吸光度的降低与人血清白蛋白浓度呈线性关系 ,线性回归方程为ΔA =0 0 0 4 6c +0 0 5 99,相关系数r=0 .995 .讨论了反应条件对实验的影响 ,对人血清中的总蛋白进行了测定 ,结果满意 。
In a Clark-Lubs buffer at pH 1.8, the color of indigo carmine(IC) dye is reduced by the interaction of human serum albumin(HSA) and IC. The linear relationship holds between the reduced absorptivity of dye and concentration ranging from 6~34 mg/L.The regression equation is ΔA=0.004 6c+0.059 9 with the correlation to be 0.995. The effects of experiment conditions are tested, and the proposed method is applied to determine the proteins in human serum with satisfactory results. The interaction mechanism of protein and dye is discussed initiatively.
