谷氨酰胺转胺酶催化羊毛接枝改性机理研究

摘要 谷氨酰胺转胺酶(MTG酶)可催化蛋白质或肽链中谷氨酰胺残基上的γ-羧酰胺基和伯胺基之间的酰基转移反应,将伯胺类物质引入蛋白质。利用MTG酶的这一催化特性,通过2,4,6-三硝基苯磺酸(TNBS)封闭羊毛中赖氨酸残基上的伯氨基,表明了谷氨酰胺转胺酶催化羊毛接枝ε-聚赖氨酸的作用机制。 Transglutaminase(MTG) could catalyze the acyl transfer reaction between the gamma carboxy amide and primary amine of the glutamyl residue groups in protein or peptides, and thus primary amine-containing substances incorporated into protein molecules. Based on the catalytic propertie of MTG, 2,4,6-trinitro-benzene-sulfonic acid(TNBS) blocked the primary amine groups in the lysine residue of wool, and the mechanisms of MTG catalyzing grafting of ε-polylysines on wool were illustrated.
