Schwann细胞转染Ha-Ras对 β_1 ,4-半乳糖基转移酶I,II,V表达的影响

摘要 目的 :研究原代Schwann细胞转染持续激活型原癌基因Ha Ras后β1,4 半乳糖基转移酶I,II ,V(β 1,4 galactosyltransferaseI ,II ,V ;β 1,4 GalT I,II,V)表达变化。方法 :构建Ha Ras表达质粒和分离纯化大鼠Schwann细胞 ,将Ha Ras表达质粒转染到Schwann细胞中。采用NorthernBlot方法分析转染细胞中Ha Ras转染情况和转染后β 1,4 GalT I,II,V的表达变化。结果 :原代Schwann细胞转染持续激活型Ha Ras后 ,β 1,4 GalT I随转染时间延长而表达增高 ,而 β 1,4 GalT II及V的表达无变化。 结论 :β 1,4 GalT I随Schwann细胞转染时间Ha Ras延长而表达增高 ,提示原代细胞表面的 β 1,4 GalT I可能与Ha Ras影响原代细胞增殖的信号途径相关。 Objective:To investigate the changes of β 1,4 galactosyltransferase I,II and V(β 1,4 GalT I,II and V) in Schwann cells transfected with oncogene Ha Ras. Methods: The constructed expression plasmids of oncogene Ha Ras were transiently tranfected into purified Schwann cells by liposomes. The changes of β 1,4 GalT I,II and V in Schwann cells transfected with the expression plasmids of Ha Ras was studied by Northern Blot. Results: The expression of β 1,4 GalT I mRNA was increased in Schwann cells transfected with Ha Ras while those of β 1,4 GalT II and V mRNA had no obvious change. Conclusion: It is suggested the β 1,4 GalT I of cell surface may associate with the path of influencing cell proliferation by oncogene Ras. \[
