摘要
目的 观测创伤性脊髓损伤后神经细胞外氨基酸含量的变化及亚低温对其的影响。方法 应用脊髓内微透析技术采集大鼠脊髓损伤区灰质细胞外液 ,采用反相高效液相色谱荧光法结合邻苯二甲醛柱前衍生技术 ,测定氨基酸类神经递质含量。结果 常温组 (A组 )和亚低温组 (B组 )损伤前细胞外氨基酸含量是稳定的 ,两组间差异无显著性 (P >0 .0 5 )。常温组脊髓损伤后细胞外氨基酸含量升高 (P <0 .0 1) ,其中尤以GABA、Glu升高为著。亚低温组胞外氨基酸含量较伤前有所升高。结论 亚低温对正常脊髓组织细胞外氨基酸释放无影响 ,能减轻脊髓损伤后细胞外Glu、Asp、Gly、Tau的过度释放 (P <0 .0 1) ,但对GABA释放无影响 (P >0 .0 5 )
Objective To observe the effects of moderate hypothermia on extracellular amino acids after the traumatic injury of the rat's spinal cord. Methods A Laminectomy of T10 was made, a probe was inserted in the gray matter and microdilysis was performed for 90 minutes before and 180 minutes after spinal cord traumatic injury. Desalinate samples were collected at intervals of 10 minutes and analyzed by high performance liquid chromatography. Results In group A there was a several-fold rise of amino acids that peaked in the first 10 minutes fraction after trauma. Group B showed a marked decrease in GLU, ASP, Gly and Tau concentration, but there was no significant difference in GABA level between two groups. Conclusions The result shaws that the moderate hypothermia can attenuate extracellular accumulation of amino acids.
