摘要
目的 :探讨基质金属蛋白酶 2、9(MMP 2、MMP 9)在鼻咽癌及其淋巴结转移灶组织中的表达、意义及与EB病毒潜伏膜蛋白 1 (LMP1 )的相关关系。方法 :应用免疫组化 (SP法 )检测LMP1和MMP 2、MMP 9在人鼻咽粘膜慢性炎(30例 )、鼻咽癌 (30例 )和鼻咽癌颈部淋巴结转移灶 (2 4例 )组织中的表达。结果 :鼻咽癌和鼻咽癌颈部淋巴结转移灶组织中LMP1和MMP 2、MMP 9的阳性率明显高于鼻咽粘膜慢性炎。在鼻咽癌及鼻咽癌颈部淋巴结转移灶标本中还发现癌巢周边的成纤维细胞、癌周浸润的单核细胞、淋巴细胞以及血管内皮细胞中也有MMP 2和MMP 9的表达。Spearman相关分析显示 ,在鼻咽癌组织中LMP1与MMP 2、MMP 9的表达无相关性 ;而在鼻咽癌颈部淋巴结转移灶组织中 ,LMP1的表达与MMP 2、MMP 9的表达呈中度正相关。结论 :MMP 2、MMP 9在鼻咽癌细胞突破基底膜向外扩散及转移中起到重要作用 ;在鼻咽癌淋巴结转移灶中LMP1与MMP 2、MMP 9的表达呈正相关 ;
Objective:To investigate the relationship of EB virus latent membrane protein 1 (LMP1)to matrix metalloproteinase 2(MMP 2) and MMP 9 in nasopharyngeal carcinoma (NPC) and lymph node metastasis.Methods:LMP1,MMP 2 and MMP 9 were detected by SP immunohistochemical technique in 30 NPCs,24 lymph node metastases and 30 specimens of chronic nasopharyngitis.Results:The expression rates of LMP1,MMP 2 and MMP 9 in NPCs and lymph node metastases were obviously higher than those in control specimens( P <0.01).Immunostaining for MMP 2 and MMP 9 was seen in stromal fibroblast,macrophages,lymphocytes and vascular endothelial cells surrounding neoplastic islands besides in tumor cells.Spearman correlation analysis showed that there was a positive relationship of LMP1 to MMP 2 and MMP 9 in lymph node metastases( P <0.01)rather than in NPCs(P>0.05).Conclusion:MMP 2 and MMP 9 play an important role in the invasion and subsequent metastasis of NPC by degrading basement membrane components,and LMP1 may be also implicated in the carcinogenesis,invasion and metastassis of NPC.
