摘要 介绍了一种新的测定半胱氨酸和还原型谷胱甘肽的荧光光度法。 N- [对 - (2 -苯并口恶唑基 ) -苯基 ]-马来酰亚胺是一种有很弱荧光的活性试剂 ,可以和硫醇定量反应 ,其加成产物有强烈荧光。利用加成产物的荧光 ,可以测定半胱氨酸和还原型谷胱甘肽 ,检出限量分别是 4 .0× 10 - 1 0 m ol· L- 1 和 3.2× 10 - 1 0 m ol· L- 1 (SNR=3)。该方法可以用于蛋白质和胱氨酸水解液中半胱氨酸的测定。 A new spectrofluorimetric method for the determination of cysteine (Cys) and reducing glutathione (GSH) has been developed. N [p (2 benzoxazole) phenyl]maleimide,which was a reactive reagent with weak fluorescence, quantitative reacted with thiols, and the products had strong fluorescence.Based on the fluorescence of the product, the concentrations of Cys and GSH could be determined. The detection limits of Cys and GSH were 4.0×10 -10 mol·L -1 and 3.2×10 -10 mol·L -1 (SNR=3) respectively. This method was applied to the determination of Cys in protein hydrolysate and cystine electrolyte with recoveries of 98.4%~104.2%.
