摘要
经诱变获得了一株高产谷胱甘肽(GSH)、多糖和麦角固醇的甲硫氨缺陷型(Met-)变株——啤酒酵母(Saccharomycescerevisiae)M05-37.该变株经培养基和培养条件优化后(蔗糖蜜为碳源,酵母粉和(NH4)2SO4为氮源,最适pH5.5~6.0,最适温度28~30℃),其活性物质的生物合成能力得到显著的提高.每100mL发酵液含GSH10.78mg,胞外多糖34.24mg,麦角固醇2.20mg,分别比出发株提高79.36%,58.07%和44.76%.
The mutant Saccharomyces cerevisiae M05-37, which was a methioninedefected strain producing a large amount of glutathione (GSH), exopolysaccharide and ergosterol, was successfully bred by mutation Its capacity to synthesize the active metabolisms including GSH, exopolysaccharide and ergosterol were great improved after its optimization for media and culture condition(sugar cane molasses as carbon resource, yeast powder and (NH4)2SO4 as nitrogen resource, at optimal pH 55~60 and optimal cultural temperature 28~30℃).The production of GSH, polysaccharide and ergosterol reached 1078 mg, 3424 mg and 22 mg per milliliter, respectively. Compared with the initial strain, the production of the above three bioactive metabolisms by the mutant were increased at 7936%, 5807% and 4736%, respectively.
