摘要
探讨了微型月季离体快速繁殖技术中的外植体表面灭菌方法,并运用二次回归正交分析法研究了不同浓度的细胞分裂素6-BA及生长素NAA对离体茎增殖的影响.试验结果表明,消毒方式采用磁力搅拌,消毒液浓度为1%,时间为15~20min,可使消毒效果达到最佳;微型月季茎段分化培养基中,激素浓度为6-BA1.93mg·L-1、NAA0.33mg·L-1时可获得最大增殖数;当培养基中6-BA的浓度控制在1.68~2.75mg·L-1范围内,NAA的浓度控制在0.07~0.54mg·L-1范围内时,接种10个芽,培养40d后,95%以上均可获得50个芽以上的增殖.
This paper discusses how to disinfect surface of explant and analyze affection of deferent concentration 6-BA and NAA on in vitro shoot propagation of miniature rose by quadratic regressive orthogonal devise in the rapid multiplication technique of miniature rose (Rosa Chinensis Minima).Draw a conclusion: We can get the best effect of disinfection when disinfection liquid concentration is 1%,time is 15~20 minute,disinfection way is magnetic force mixing,and we can get the best the buds proliferation when combination of hormone concentration in shoot differentiation medium of miniature rose are 6-BA 1.93?mg·L-1,NAA 0.33?mg·L-1.While the 6-BA concentration was controlled in range of 1.68~2.75?mg·L-1 and NAA concentration was controlled in range of 0.07~0.54?mg·L-1,we can have 95% chance to get more than 50 buds after 10 buds was inoculated and cultured for 40 days.
