摘要
大量实验表明,TNFα可特异性杀伤肿瘤细胞,但并非所有癌细胞对TNFα都敏感。本实验仅就重组人TNFα及其与不同抗癌药物联合应用对人食管鳞状上皮癌细胞系(109细胞)进行细胞毒性观察。在5×10~5/ml 109细胞中,rh-TNFα单用或加入不同浓度的5Fu和平阳霉素合用,培养24小时后用MTT法测定细胞毒性,结果证明:TNFα单用对109细胞有一定的细胞毒作用;与5Fu和平阳霉素合用时,rh-TNFα在同等剂量下,细胞毒作用明显增强,表明109细胞属于对TNFα低度敏感的细胞系;TNFα单用虽可杀伤肿瘤细胞,但效果不甚理想,当与抗癌药物,尤其是两种以上的抗癌药合用时,对肿瘤细胞的毒性呈显著协同作用。
With use of recombined human tumor necrosis factor alpha (rh-TNFα), 5-fluorouracil (5Fu) and bleomycin, cytotoxic effect on 109 cell line was observed. Different concentration of these drugs (TNF: 5,1,0.2,0.04μg/ml; 5Fu: 2500,500,100,20pμg/ ml; bleomycint 500,100,20,4μg/ml) were added into 5×10~5/ml 109 cells. After culture for 24 hours, cytotoxicity were detected with MTT method. The results showed that rh-TNF alone has a certain cytotoxic effect on 109 cell, and the cytotoxicity has been enhanced when TNF combined with 5Fu or bleomycin as above concentration(even half), especially in combination of the three drugs. But, as the concentration of TNF Increasing, there was no obviously increment in cytotoxicity.The results suggested that 109 cell was a low sensitive to rh-TNF. But TNF combined with antitumor drugs, particularly more than two kinds of the drugs, its antitumor effect was enhanced significantly.
