酶标SpA法在检测小鼠同系抗独特型中的应用

Detection of Anti-idiotypic Antibody Produced in Syngenic Mouse by HRP-SpA

本研究用辣根过氧化物酶标记的葡萄球菌蛋白A(HRP-SpA)检测同系小鼠抗独特型抗体。主要步骤为:用以低亲合力结合SpA的小鼠免疫球蛋白如IgG1包被,加入待检抗独特型抗体,最后以HRP-SpA显示其反应。本法简便、灵敏、特异性好,而且可用于对抗原结合部位特异性与非特异性独特型的鉴别。

In this work, HRP-SpA was used to detect anti-idiotypic antibody produced in syngenic mouse. The main procedures were as follows:coating by mouse IgG subclass with low affinity to SpA such as IgG1 or IgM, adding anti-idiotype, then HRP-SpA was used to display the binding between idiotype and anti-idiotype. This method is simple, sensitive and specific, it can be used to analyze the anti-idiotypic antibody against the antigen binding specific or nonspecific sites of idiotype.
