The Preparation, Identification and immunoprotection of Anti-idiotypic Antibodies against STM H and O Antigen

摘要 用纯化的鼠伤寒沙门氏菌(Salmonella Typhimurium STM)H和O抗原的抗独特型(idiotype Id)抗体免疫成年和新生BALB/c小鼠,再用20和10LD_(50)STM活菌攻击,观察抗Id对小鼠的免疫保护作用。以3日内存活数为保护率指标。成鼠组结果是Ab2-O和其F(ab’)_2分别为62.6%和71.4%;Ab2-H和其F(ab’)_2均为12.5%;Ab2-O+Ab2-H为75%;STM死菌苗为100%。新生鼠组Ab2-O为80%;Ab1-O+Ab2-O为66.7%;Ab1-O为57.1%;Ab2-H、Ab1-H+Ab2-H和STM死菌苗均无保护作用。Ab2免疫的乳鼠和家兔血清中均查出抗STM O和H抗原的抗体。鉴于O抗原是沙门氏菌保护性抗原,新生乳鼠对这类TI抗原缺乏免疫应答能力,而抗Id作为TD抗原,则能诱导这些动物产生免疫应答。故设想模拟多糖的抗Id可作为疫苗用于这类个体的免疫预防。 The immunoprotection of anti-idiotypic antibodies against STM (Salmonella Typhimurium) for adult and neonate BALB/c mice was studied. The mice were immunized with different anti-idiotypic antibodies and STM vaccine. Ten days after last immunizing they were challenged with 20 and 10 LD50 living STM respectively. The immunoprotection is expressed by the survival of mice on 3 days after challenge. The results show that the rate of protection was 62.6% and 71.4% with Ab2-O and its' F(ab')2; 12.5% with both Ab2-H and its' F(ab')2; 75% with Ab2-H + Ab2-O and 100% with inactive STM vaccine for adult mice; 80% with Ab2-O, 66.7% with Ab2-O + Ab1-O, 57.1% with Ab1-O and nonprotection with Ab2-H and STM vaccine for neonate mice. It is known the protective antigen of salmonella is o antigen. These antigens are TI-antigen and are difficult to induce the immunoresponse in neonate mice. Whereas the anti-idiotypic antibodies are TD-antigen, so it can induce such immunity in these animals. The antibodies against H and O antigens of STM were detected from both rabbits and mice immunized by Ab2.The results are shown that the anti-idiotypic antibodies which mimic polysacharids antigen might be used as a vaccine for neonates.
