摘要
用神经氨酸酶及羧肽酶B处理血浆,依次经C4高压琼脂糖电泳、免疫固定、薄层激光扫描等步骤,并结合血浆免疫火箭电泳定量法对我国武汉地区随机人群的血浆C4总量及C4两种同种型,主要别型,单、双QO,重复座位等时的血浆C4含量进行了检测与分析。结果发现:遗传因素明显影响血浆C4水平,因而在群体中表现宽广的C4浓度范围,特别在有C4QO与C4重复基因座位时差异特别明显。这一研究提示,不同的C4基因型,循环C4含量表现明显差异。临床上检测C4含量时应该考虑患者的C4基因型别。
Blood C4 level in normal random population and in subjects with different known C4 isotypes,main C4 allotypes,with or without C4QO and duplicated C4 genes in the Han population of Hubei district was determined with high volgage agarose electrophoresis,immunofixation.thin layer laser scanning and immunorocket electrphoresis. The results clearly showed that the genetic factors influence plasma C4 concentration and contribute to the wide variation seen in population. The mean total plasma C4 level in normal random population was 474mg/L,in subjects getting ride of null genes (QO) was 543mg/L ,of C4A carrying persons was 234mg/L,of C4B carrying persons was 268mg/L,of C4A3 carrying persons 138mg/L籵f C4A4 carrying persons 104mg/L,of C4B1 carrying persons 124mg/L,of C4B2 carrying persons 149mg/L,of C4B5 carrying persons 193mg/L,of persons with single null gene 426mg/L,of persons with double null genes 357mg/L. When C4 gene was duplicated, the blood C4 level increased. We have determined the C4 level in four persons with C4B duplication,the C4A/C4B ratios were 1. 0 vs 1. 6 - 1. 9. This study suggested that different genotypes are likely to show different levels of circulating C4. Any clinic interpretation of blood C4 concentrations must take the C4 genotypes into account.
