两个凡纳滨对虾亲本群体的RAPD研究 被引量:10

摘要 采用Operon公司的 16个引物对两批凡纳滨对虾亲虾的基因组DNA多态性进行了检测 ,结果表明 ,第一个亲本群体对 16个引物共检测到 78个位点 ,其中多态位点数为 4 8个 ,占 6 1.5 4% ;第二个亲本群体共获得 97个位点 ,其中多态位点数为 4 9个 ,占 5 0 .5 2 %。单个引物获得的标记数为 1~ 8个。第一个亲本群体个体间的平均遗传距离为 0 .196 0± 0 .0 392 ,第二个亲本群体个体间的平均遗传距离为 0 .0 92 2± 0 .0 189。两个亲本群体间的多态位点比例为 6 6 .98% ,遗传距离为0 345 5± 0 .0 795。在OPF 0 9、OPV 19、OPZ 113个引物的扩增结果中 ,发现两个亲本群体有明显差异。 Two groups of primary parents of Litopenaeus vannamei from Hawaii have been collected for this experiment. Random amplified polymorphic DNA (RAPD) technique was employed to detect the genetic variation of two groups of primary parent shrimp from the population. A total 140 random primers were evaluated on their usefulness for shrimps. 16 primers were useful according to the quantity, quality and reproducibility of loci. With the 16 primers there are 78 loci for the first group of shrimp, 48 of which are polymorphic and the proportion of polymorphic loci is 61.54%; and for the second group there are 97 loci, 49 of which are polymorphic and the proportion of polymorphic loci is 50.52%. The average genetic distance of the first group is 0.196 0±0.039 2, whereas 0.092 2±0.018 9 of the second group. The proportion of polymorphic loci and the genetic distance is 66.98% and 0.654 6±0.079 5 respectively between two groups of parents shrimps. As a result, we can find different bands in the two parent amplified results which amplified with primers OPF 09,OPV 19 and OPZ 11. There are some differences between two groups of primary parents.
