阿苯达唑和甲苯咪唑对旋毛虫寄生肌肉的组织学与组织化学观察

Histological and histochemical observation on the effect of albendazole and mebendazole against parasitical muscle of Trichinella Spiralis

目的 观察阿苯达唑和甲苯咪唑对旋毛虫寄生肌肉的影响。方法 于感染后 10d、4 0d ,分别给感染沙鼠喂饲阿苯达唑或甲苯咪唑 30mg/kg/d ,qd ,连续 5d。同时设对照组。在感染后 2 0d、5 0d处死沙鼠 ,取腓肠肌制作冰冻切片。用组织学和组织化学方法 ,观察腓肠肌形态及ATPase、NADH TR、LDH等酶的变化。结果 旋毛虫感染沙鼠后 ,虫体周围肌纤维肿胀、变性、坏死 ,幼虫周围有炎细胞浸润 ;两治疗组肌纤维形态与相应对照组无明显改变 ;两治疗组肌肉组织ATPase、NADH TR活性均高于相应对照组 ,而LDH活性低于对照组 ;在囊包形成期 ,甲苯咪唑治疗旋毛虫感染沙鼠后其寄生肌肉ATPase、LDH活性改变较阿苯达唑明显。结论 阿苯达唑和甲苯咪唑治疗受染沙鼠后 ,肌肉组织有氧代谢增强 ,而无氧糖酵解减弱 ;在囊包形成期甲苯咪唑的治疗效果优于阿苯达唑。

Objective To study the action of albendazole(ABZ) and mebendazole(MBZ) on gastrocnemius of Mongolian jirds infected Trichinella spiralis . Methods After infected 10d 、40d, the jirds of infected groups were treated by ABZ or MBZ 30mg/kg/d for 5 day, respectively. Control groups was established. The jirds were killed after infected 20d 、50d, and taken gastrocnemius to make section frozen. The form and change of ATPase、 NADH-TR、LDH of gastrocnemius were observed by histological and histochemical methods. Results In infected gastrocnemius, edema, degeneration and necrosis were observed. The larvae of Trichinella spiralis were surrounded by inflammatory cells. The forms of gastrocnemius were had not obvious difference between treated groups and control groups. Activities of ATPase and NADH-TR in two treated groups were increased, Activities of LDH was decreased.Activities of ATPase、LDH in MBZ treated group were higher than in ABZ treated group for forming cyst period. Conclusions ABZ and MBZ can enhance aerobic metabolism of muscle, and restrict anaerobic metabolism. Effect of mebendazole was better than albendazole in forming cyst period.
