ABC法测定CMV IgM中关键试验影响因素的研究

Investigation on Various Affecting Factors of Detecting Cytomegalovirus IgM by The Method of ABC-ELISA

用正交试验统计方法,考察ABC法检测CMV IgM中关键试验因素。依据影响程度对抗原浓度、载体处理、封板液和包被时温等进行排序。结果表明载体预处理和封板液选择在ABC法试验中至关重要。采用正交试验得出的最佳试验方法较常规方法科学、准确和省时。

Using the orthogonal test, we investigated the key affecting factors of detecting cytomegalovirus IgM by ABC-ELISA. This article evaluted the affecting sequence and degree of the pretreatment of antigen and microelisa plates, the blocking solutions,as well as times and temperatures of coating antigen. The result shows that the blocking erquid and antigen pretreatment are very important in ABC-ELISA. The most condition of test chosen from the orthogonal is superior to the routine methods for determining CMV IgM, with the advan-tange of fewer time and quicker and more exact.
