摘要
以假单胞菌B5菌株 (实验室筛选 )为实验菌株 ,完成了游离细胞发酵条件优化和发酵保护剂甘油最适用量的研究。在 6g/L的苯甲酸钠和 1 1g/L培养基上 30℃培养 2 4h ,邻苯二酚 (catechol)产量达到 2 5g/L ,分子水平转化率为 5 2 3%;同时进行细胞固定化材料、固定化条件和固定化发酵条件的实验。以 1 5 %壳聚糖和 0 1%的海藻酸钙为固定化介质 ,0 1%的戊二醛交联制备得到的固定化细胞成球形 ,直径约为 2 5mm ,在苯甲酸钠 (6g/L)培养基中可连续批次发酵 12次 ,邻苯二酚平均产量约为每批次 1 5g/L。
The primary study was focused on the optimization of fermentation parameters with free cells and the most convenient dose of glycerol was completed on the base of B5 strain. Psuedomonas sp.B5 can produce catechol (2.5?g/L) from the medium containing sodium benzoate (6?g/L) and glycerol (1.1?g/L) after 24?h culture at 30?℃, and the conversion rate is 52.3% on a molar basis. At the same time, by comparison of three immobilization materials including gelatin, calcium alginate and chitosan, a good immobilization condition was established including 1.5% chitoson, 0.1% calcium alginate and 0.1% glutaraldehyde at 35?℃ for 8?h. The average diameter of immobilized cells was about 2.5?mm. For the medium which contained sodium benzoate (6?g/L), it could be fermented continuously 12 times and the average output of catechol was 1.5?g/L.
