摘要
目的 观察移植细胞在骨折愈合不同时相的存活及转归 ,及血管内皮生长因子 (VEGF)动态表达的生物学意义。方法 建立 SD大鼠老龄骨质疏松骨折的动物模型 ,将 SD雄性乳鼠颅骨体外培养的成骨细胞移植到 SD雌性鼠骨质疏松性骨折部位 ,用原位杂交及免疫组化检测骨折愈合不同时相的标本 :(1 ) Y染色体阳性表达特点 ;(2 ) VEGF、VEGF m RNA的动态表达。结果 实验组成骨细胞移植后可在受体骨折部位局部生存 ,生长繁殖 ,促进骨质疏松性骨折愈合 ,并于移植 84d后仍存活。实验组 VEGF、VEGF m RNA均在 7d左右可见有阳性表达的细胞 ,1 4 d有分泌高峰 ,其中以软骨细胞中阳性最强 ,2 8d分泌量开始下降 ,56 d后基本消失。对照组未见明显分泌高峰。结论 同种异体成骨细胞移植到老年大鼠骨质疏松性骨折部位 ,参与骨折修复的各个环节 ,可至少存活 84d。 VEGF的动态表达对促进老年骨质疏松性骨折愈合具有重要意义。
Objective Study on the follow up observation of the long term survival of the osteoblasts by Y chromosome and the biological function and behavior of VEGF duing fracture healing of osteoporosis rats Methods Set up the fracture models of osteoporosis rats and do osteoblasts grafts; as well as detect during different periods in the process of fracture healing by using immunhistochemistry to detect the expression of VEGF and in situ hybridization to study the expression of Y chromosome, VEGFmRNA, also using image analysis to deal with it Results In experimental group, Osteoblasts can survive, even proliferate in fracture areas; Osteoblasts grafts can accelerate bone fracture healing of osteoporosis rats, and still survive on 84 days after grafts In experimental group, VEGF can be seen positive after 7 days of after osteoblast grafts, and there have the highest quantities in chondrocytes about 14 days of post transplantation Then began to decline after 28 days, and disappear after 56 days However there are not obviously high quantities in control Conclusions Osteoblasts grafts have a long term survival and enhance bone fracture healing of osteoporosis rats VEGF plays a very important role in accelerating fracture healing of osteoporosis rats
