电针对脊髓损伤早期bcl-2 mRNA及蛋白表达的影响 被引量:24

摘要 目的 :探讨电针治疗脊髓损伤的机制。方法 :成年雄性SD大鼠 ,随机分为模型 (MC)组、电针 (EA)治疗组、甲基强的松龙 (MP)组及假手术 (SO)组。采用改良的Allen s捶击法致大鼠T10 脊髓损伤 ,应用原位杂交和免疫组织化学方法及图像定量分析法观察脊髓损伤早期bcl 2mRNA和蛋白的表达。结果 :MC组脊髓损伤后 6hbcl 2mRNA表达有增高趋势 ,伤后 2 4h表达增高 ;伤后6hbcl 2蛋白表达未见明显变化 ,伤后 2 4h表达有增高趋势。EA组bcl 2mRNA及蛋白表达均高于模型组 ,与MP组相比差异无显著性意义。结论 :电针可上调bcl 2mRNA及蛋白的表达 ,从而抑制细胞凋亡 。 Objective To explore the mechanism of electroacupuncture for treatment of spinal cord injury. Methods Adult male SD rats were divided into model control group, electroacupuncture group, methylprednisolone group and sham operation group. Injury of T 10 spinal cord was induced by modified Allens method and the expressions of bcl 2 mRNA and protein were observed by in situ hybridization, immunohistochemical analysis and image quantitative analysis. Results In the model group, expressions of bcl 2 mRNA tended to increase 6 h after injury of the spinal cord and increased 24 h after the injury; expression of bcl 2 protein did not change 6 h after the injury and tended to increase 24 h after the injury. Expressions of bcl 2 mRNA and protein increased in the electroacupuncture group as compared with the model group, with no significant difference as compared with the methylprednisolone group. Conclusion Electroacupuncture can up regulate the expressions of bcl 2 mRNA and protein, so as to inhibit apoptosis and protect nervous cells in early spinal cord injury.
