核黄素预处理对缺血再灌注损伤的心肌线粒体保护作用的研究 被引量:3

摘要 目的 探讨核黄素对缺血再灌注损伤心肌线粒体的保护作用。方法 实验用家兔随机分成 4组 :假手术组(SC) ,缺血再灌注损伤组 (IR) ,缺血预处理组 (IPC) ,核黄素预处理组 (RBF) ;4组均于再灌注 30min后立即取结扎线下部位约 0 .5cm处心肌组织 1g ,制备线粒体悬液之后检测SDH活力、MDA含量及GSH PX活力。 结果 与SC组比较 ,结扎后IR组各时点的收缩及舒张功能明显下降 (P <0 .0 5 ) ,MDA含量显著升高 (P <0 .0 5 ) ,GSH PX活力则显著降低 (P <0 .0 5 ) ,SDH的活力非常显著地下降 (P <0 .0 5 )。与IR组比较 ,RBF组和IPC组各时点的收缩及舒张功能下降程度明显减轻 (P <0 .0 1,P <0 .0 5 ) ,MDA含量显著降低 (P <0 .0 5 ) ,GSH PX活力显著升高 (P <0 .0 5 ) ,SDH活力明显升高 (P <0 .0 1,P <0 .0 5 )。且RBF组SDH活力明显高于IPC组 (P <0 .0 5 )及SC组(P <0 .0 5 )。结论 核黄素保护组能较明显地抑制缺血再灌注损伤时线粒体脂质过氧化作用 ,保护心肌细胞线粒体的结构和功能 ,对再灌注引起的损伤起到保护作用。 Objective\ To study the protective effects of riboflavin preconditioning on myocardial ischemia reperfusion injury . Methods\ 36 rabbits,weighing 1.8~2.0 kg,were randomly divided into four groups:sham operation control group(SC,n=9),ischemia repurfusion injury group(IR,n=9),ischemia preconditioning group(IPC,n=9),riboflavin preconditioning group(RBF,n=9).The model of myocardial ischemia reperfusion injury was produced by coronary artery ligation.The systolic and diastolic function of left ventricle was measured .The activity of SDH and GSH PX and the content of MDA were observed respectively.Results\ Compared with sham operation control group(SC) ,riboflavin preconditioning group(RBF) obviously increased ±dp/dtmax and LVSP ,markedly decreased LVEDP(P<0.05).As compared with ischemia reperfusion group(IR),ischemia preconditioning group(IPC) and riboflavin preconditioning group(RBF) showed significantly increased activity of SDH and GSH PX and the content of MDA (P<0.05) .Conclusion\ Riboflavin preconditioning has notable protective effects on myocardial ischemia reperfusion injury and can improve the LV function.
