脑胶质瘤基因治疗中丙氧鸟苷对细胞系的毒性分析

摘要 目的 :研究丙氧鸟苷 ( GCV)对 C6BPBAG脑胶质瘤细胞、psi2细胞、psi2 STK细胞的毒性。方法 :将 C6BPBAG脑胶质瘤细胞、psi2细胞、psi2 STK细胞分别在不同浓度 GCV的培养基中培养 ,观察以上 3类细胞对 GCV的毒性反应。结果 :在有效浓度 ( 0 .0 1~ 1 0 μmol·L-1 )中 ,GCV对C6BPBAG脑胶质瘤细胞、psi2细胞几乎无伤害 ,而经改造后的 psi2 STK细胞对 GCV毒性明显提高。结论 :通过体外实验证实由逆转录病毒载体 ( STK)传导的 HSV- TK基因 ,在培养的 C6胶质瘤细胞中 ,可提高 GCV对 psi2 Objective: To study the toxic effects of Ganciclovir(GCV) on C 6BPBAG.psi2 and psi2STK. Methods: In order to observe the toxic effects of GCV on C 6BPBAG,Psi2 and Psi2STK, these cells were planted at a density of 10 4 cells per cm 2 with different concentration of GCV. Results:When the density of GCV was 0.01~10 μmol·L -1 ,GCV was almost not harmful to C 6BPBAG and psi2. However when psi2STK was transformed, the toxic effects of GCV on Psi2STK were increased singnificantly. Conclusion: According experiment in vitro , it is proved that when HSV TK gene is transferred by STK, the sensitivity of GCV to Psi2STK is improved.
