摘要
以对药物的抗性和敏感性为遗传标记,筛选掘氏疫霉(Phytophthora drechsleri Tucker)双标记菌株为配对亲本.测定了采集自我国江苏、广东、上海及来自美国菌种保藏委员会的28个掘氏疫霉野生型菌株对36种药物的抗性,筛选出2个双标记配对组合:Phy607×Phy112、Phy115×Phy64,其中,菌株Phy607(A_1)、Phy64(A_2)抗放线菌酮对新霉索敏感,Phy115(A-1)、Phy112(A_2)抗新霉素对放线菌酮敏感.上述配对组合可用于掘氏疫霉有性生殖行为的研究.上述菌株对放线菌酮和新霉素的抗性由细胞核杂合基因控制,抗性为显性遗传.测定结果还表明,掘氏疫霉种内菌株间对甲霜灵、恶霉灵、乙磷铝、金霉素、稻瘟索等5种药物的抗性存在明显差异,可能作为该种遗传研究的标记性状.
The selection of mating pairs coming from parents with double-marker(resistant and sensi-tive to drugs)from wild type isolates of Phytophthora drechsleri is reported in this paper.Two mating pairs ofparents with double-marker,Phy607xPhy112 and Phy115xPhy64 were selected,in which,isolates Phy607(A_1 and Phy64(A_2)were resistant to actidione and sensitive to neomycin.On contrary,Phy115(A_1)and Phy112(A_2)were resistant to neomycin and sensitive to actidione.Those two mating pairs could be used for theresearch of behavior in sexual reproduction of Phytophthora drechsleri.Resistance to actidione and neomycinwas inherited dominant and might be controlled by heterozygote gene(s)in nuclei.Results also showed that,resistance to other 5 drugs including metalaxyl,hymexozal,phosetyl AL,aureomycin and blastitin-s wasobviously different among isolates of P.drechsleri,and was possible to be selected as marker in genetical re-searches.
