摘要
从患关节炎病鸡肠道组织内分离到一株禽呼肠孤病毒(ARV),定名为 NAU-8902.该病毒能在鸡胚卵黄囊内繁殖并致死鸡胚.鸡胚毒适应鸡胚肝细胞(CELi)和鸡胚成纤维细胞(CEF)后,产生以合胞体为特征的细胞病变,分离毒 CEF-2代的 TCID_(50)为10^(-7.39)/0.1ml.电镜观察,病毒具有双层衣壳,20面体对称,直径50~65nm,无囊膜.血清学试验证明,病鸡体内含有禽呼肠孤病毒抗体,聚丙烯酰胺凝胶电泳结果显示,病毒基因组由10个节段组成,带形与参考株(ARV FDO 株)相似,呈3-3-1-3分布.将适应细胞后的分离毒回归1日龄 SPF 雏鸡,复制出病毒性关节炎.结果表明,先将病料接种于鸡胚卵黄囊,然后适应 CELi,最后在 CEF 细胞上传代,是一种适宜的分离 ARV 的方法.
A strain of avian reovirus(ARV)designated NAU-8902 was isolated from a chicken witharthritis.The virus could replicate in chicken embryo yolk sac(YS),resulting in embryonic death.After thevirus was adapted to the chicken embryo liver cell(CELi)culture and chicken embryo fibroblast(CEF)cul-ture,cytopathic effect(CPE)characterized by the formation of syncytia was induced with 10^(-7.39)TCID50/0.1 ml in CEF-2.Under electron microscope,the virus particles(50~65 nm in diameter)are non-enve-loped and have a double capsid structure of icosahedral symmetry.The viral genome was composed of tensegments with the migration pattern of 3-3-1-3,similar to strain FDO,as determined by PolyacrylamideGel Electrophoresis(PAGE).After NAU-8902 was inoculated into l-day-old SPF chicken,arthritis wasreproduced.
