荔枝核增强2型糖尿病-胰岛素抵抗大鼠胰岛素敏感性作用 被引量:58

摘要 目的 :观察荔枝核水提取物 (以下简称荔枝核 )对 2型糖尿病 (T2DM )大鼠胰岛素抵抗 (IR)的影响 ,并初步探讨其作用机制。方法 :采用给大鼠腹腔一次性注射小剂量链脲霉素 (30mg·kg-1) ,并加饲高热量饮食 (富含脂肪和蔗糖 ) ,制备T2DM伴IR大鼠模型 ,观察荔枝核对病鼠血糖、血脂、胰岛素和胰岛素敏感性指数的影响。结果 :荔枝核能显著降低T2DM伴IR模型大鼠空腹血糖和口服葡萄糖耐量试验后 2h血糖 (P <0 0 5 ) ,同时改善了病鼠的糖耐量减退。荔枝核 3个剂量组还能明显降低病鼠的血清总胆固醇 (TC)和三酰甘油 (TG) (P <0 0 1) ,提高高密度脂蛋白 胆固醇 (HDL C)含量和HDL C/TC比值 (P <0 0 1)。降低病鼠高胰岛素血症 ,提高了胰岛素敏感性指数 (P <0 0 1)。实验中未发现明显的不良反应。结论 :荔枝核对T2DM伴IR大鼠有降血糖及调血脂的作用 ,并改善其IR。 Objective:To investigate the effects of Litchi seed water extractant(LS)on insulin sensitivity in type 2 diabetic rats with insulin-resistance (IR) and the primary pharmacological mechanism.Methods:The model for type 2 diabetic-insulin-resistant rats (T2DIRR) were performed through intraperitonea injection of lower dosage of streptozocin(30mg·kg -1) and high caloric diet (abounded with sugar & fat),the effects of LS on blood and/or serum contents of glucose,lipids,insulin and insulin sensitive index(ISI) were observed in the model rats.Results:LS lowered significantly fasting blood glucose (FBG) and 2-hours blood glucose (2hBG) after oral glucose tolerance test (OGTT) (P<0.05) and improved impaired glucose tolerance (IGT);decreased markedly the levels of total cholesterols (TC) and triglyceride (TG) (P<0.01),and increased the content of high-density lipoprotein cholesterol (HDL-C) and ratio of HDL-C/TC (P<0.01);and reduced the concentration of insulin in hyperinsulinemia,and potentiated ISI (P<0.01) in T2DIRR.Any noxious and side effects didn't occur in the experiment.Conclusion:In the experiment with type 2 diabetic-insulin-resistant rats,LS can reduce level of blood glucose,regulate components and content of blood lipids,and improve insulin sensitivity.
