摘要 目的:探讨白血病抑制因子(Leukemia Inhibitory Factor,LIF)在实验性骨性关节炎(osteoarthritis,OA)不同时期的表达变化。阐明LIF在OA发病学中的意义。方法:设右侧膝为实验侧,采用Colombo兔实验性骨性关节炎模型;左侧膝行假手术为对照侧。于不同时间处死动物,取膝关节滑膜和软骨组织。采用光镜观察骨性关节炎不同阶段的组织学改变,以免疫组织化学技术检测LIF关节软骨和滑膜中的表达变化。结果:实验组术后42d光镜显示关节滑膜及软骨明显的OA组织学改变;实验组关节软骨细胞LIF的表达量明显高于对照组(P<0.05;术后28d软骨细胞中LIF的表达明显高于其他时间点(P<0.05),不同时间点实验组与对照组之间对应比较均有显著性差异(P<0.05);实验组不同时间段滑膜细胞能不同程度的表达LIF,对照组关节滑膜细胞未见LIF表达。结论:LIF在OA软骨和滑膜细胞中的异常表达及变化具有特异性,其原因与OA不同时期软骨和滑膜细胞变性有直接关系。LIF在OA发病中起重要作用,是OA发生发展的重要介导因素之一。 Objective: To study the role and expression of Leukemia Inhibitory factor(LIF) in experimental osteoarthritis. Methods: Operation was performed in the right knee of each rabbit, and the sham operation was performed in the left knee, cartilage and synovium of knee were collected to observe the histological changes of osteoarthritis in different time points and the expression of LIF in the cartilage. And the synovium were detected by immunohistochemical analysis at the same time. Results: The typical degenerative change of cartilage and synovium were detected histologically at 42 days after operation. LIF expressed differently in cartilage of experimental group in different time points. In 28 days after operation, the expression of LIF was increased dramatically compared to any other time point (P < 0.05). There was significant difference of expression between the operating group and the sham operation group at the same time point. (P<0.05). The expression of LIF was shown in the synovium of experimental group but not in the control group. Conclusion: The results indicate that LIF is overexpressed singularly in osteoarthritic cartilage and synovium at different time points, which is considered to correlate with the degeneration of chondrocytes and synoviocytes. LIF is one of the important mediators that can contribute to the pathogenesis of OA.
