摘要
采用心钠素放射免疫测定法对体外培养的新生大鼠心房和心室肌细胞心钠素的分泌进行了动态观察,并以免疫组织化学方法对心肌细胞内心钠素的定位和含量进行了探讨。结果表明:(1)培养于无血清培养基中的心房和心室肌细胞,在一定时限内以恒定的分泌率分泌心钠素。用此模型研究多种因素对心钠素分泌的影响具有科学性和可行性。(2)培养的新生大鼠心室肌细胞与心房肌细胞相比,虽然前者含有较少的心钠素分泌颗粒,但两者却具有相同的分泌率,提示两型细胞的心钠素分泌机制可能存在差异。
The cecretion of atrial natriuretic peptide(ANP)from cultured neonatal rat atrial and ventricular myocytes was observed with specific radioimmunoassay and immunocytochemical analysis.The results indicated that 1.The rate of ANP secretion from cultured neonatal rat cardiocytes in serum-free medium was consistently maintained within a limited time.The established culture model of neonatal rat atrial or ventric- ular myocytes is scientific and feasible in studying the factors that affect secretion of ANP in vitro.2.neonatal rat ventricular myocytes in culture,although contained fewer ANP secretory granules,seereted substantial amounts of ANP at a comparable level to that of atrial myocytes,The cellular mechanisms of ANP secretion probably differ in the two cell types.
