摘要
作者提出了反相高效液相色谱法测定新生儿血清中未结合胆红素的方法。在咖啡因试剂存在下血清中未结合胆红素与白蛋白解离被定量地萃取到氯仿中,使用C_(18)柱的反相高效液相色谱法在450nm 处监测胆红素。以美国国家标准局 SRM916a 胆红素为原始标准,胆红素加入血清后的平均回收率为0.996(s=0.018,n=16)该方法被建议为新生儿血浆未结合胆红素的标准参考分析法。
A high performance liquid chromatographic(HPLC)method for the analysis of unconjugated bilirubin in neonatal serum is presented.Bilirubin was dissociated from protein with caffeine reagent and extracted with chloroform.An isocratic reversed-phase HPLC system based on a C_(18) column was used.Bilirubin was detec- ted at 450nm.Bilirubin SRM 916 a from National Instiutute of Standa(?)ds and Tech- nology USA was the primary callbrator.An average recovery factor of 0.996(s=0.018; n=16)was obtained for bilirubin added to neonatal cord sera.The measurement range extended from (?)5 to 500 μmol/L.Itis proposed as a reference method for uncon- jugated bilirubin in neonatal sera.
