摘要
目的 研究碱性成纤维细胞生长因子 (bFGF)对辐射诱导的血管内皮细胞凋亡的作用及其信号转导途径。方法 以6 0 Coγ射线照射建立原代分离培养的人脐带静脉内皮细胞凋亡模型 ,以流式细胞术 (Annexin V FITC +PI标记 )评价细胞凋亡率。观察不同浓度的bFGF和MEK的抑制剂U0 12 6对细胞凋亡的影响。结果 bFGF对辐射诱导的脐静脉内皮细胞凋亡有明显的抑制作用 ,细胞凋亡率可从 (35 2 2± 5 5 ) %降至 (11 6 7± 3 8) % ,而U0 12 6可部分阻断bFGF的抗凋亡作用 ,细胞凋亡率由 (10 5 9± 1 6 ) %升至 (17 96± 1 4 ) %。结论 bFGF可能通过MAPK信号转导途径抑制辐射诱导的脐静脉内皮细胞凋亡。
Objective To study the effect of bFGF on radiation-induced apoptosis in vascular endothelial cells and its signal transduction pathways. Methods Primary cultured of HUVEC (human umbilical vein endothelial cells) was irradiated with 60 Co γ-rays to establish a cell apoptosis model. Flow cytometry with Annexin-V- FITC+ PI labeling was used to evaluate cell apoptosis. Different amounts of bFGF and MEK inhibitor U0126 were used to study their effects on radiation-induced endothelial cell apoptosis. Results We found that bFGF could inhibit radiation-induced apoptosis in HUVEC, i.e., the apoptotic rate decreased from (35\^22±5\^5)% to (11\^67±3\^8)%.Meanwhile,U0126 could partly block the anti-apoptosis effect of bFGF,the apoptotic rate increased from (10\^59±1\^6)% to (17\^96±1\^4)%. Conclusion MAPK signal transduction pathway may be involved in the inhibitory effect of bFGF on radiation-induced apoptosis in HUVEC.
