摘要
目的 研究ets 1反义寡核苷酸对胃癌细胞株SGC 790 1侵袭力的影响。方法 应用ets 1反义、正义寡核苷酸转染SGC 790 1细胞 ,并设磷酸盐缓冲液 (PBS)对照组。转染后应用逆转录 聚合酶链反应 (RT PCR)检测ets 1mRNA的变化 ,应用Transwell小室检测体外侵袭力的变化 ,并应用裸鼠胃癌皮下转移模型检测对体内侵袭力的影响。结果 转染ets 1反义寡核苷酸后 ,ets 1mRNA表达降低 (0 .42 3± 0 .0 86vs 0 .815± 0 .176、0 .80 7± 0 .169,P <0 .0 1) ,侵过小室滤膜细胞数目减少 (97.1± 11.1vs 198.7± 18.3、2 0 5 .8± 2 2 .2 ,P <0 .0 1) ,裸鼠胃癌生长明显受到抑制 (1.2 8±0 .13vs 1.18± 0 .11、0 .60± 0 .11,P <0 .0 1)。结论 ets 1反义寡核苷酸能够抑制胃癌细胞株SGC 790
Objective To investigate the effect of ets-1 antisense oligoxydeonucleotide (AsODN) on invasive ability of gastric cancer cell line SGC-7901.Methods Cultured cells were divided into 3 groups: control group,sense oligoxydeonucleotide (sODN) group and AsODN group.After transfection for 24 h,cultured cells were harvested.The changes of ets-1 mRNA were detected by the reverse transcription polymerase chain reaction method (RT-PCR) and invasive ability was measured by the number of cells to penetrate polycarbonates coated with matrigel.Human gastric carcinoma transplanted subcutaneously in nude mice was constructed to detect the effect of est-1 AsODN on invasive ability in vivo.Results ets-1 AsODN could down-regulate the expression of ets-1 mRNA (0.423±0.086 vs 0.815± 0.176,0.807±0.169,P<0.01).The number of cells that penetrated polycarbonates was significantly lower in the AsODN group than sODN group and control group (97.1±11.1 vs 198.7±18.3,205.8±22.2,P<0.01).ets-1 AsODN could inhibit the growth of gastric carcinoma transplanted in nude mice (1.28±0.13 vs 1.18±0.11,0.60±0.11,P<0.01).Conclusion ets-1 AsODN can inhibit the invasive ability of gastric cancer cells.
