阻断转化生长因子-β1自分泌环对骨肉瘤细胞生物学活性和化疗药物敏感性的影响 被引量:3

Effects of TGF-β1 autocrine blockage on the osteosarcoma cells

摘要 目的 观察阻断转化生长因子 β1(TGF β1)自分泌环对肿瘤细胞增殖活性和化疗药物敏感性的影响。方法 将反义TGFβ1基因转染骨肉瘤细胞MG 63 ,采用四甲基偶氮唑蓝微量酶反应比色法 (MTT)检测细胞增殖活性。以含 0、0 .2 5、0 .5 0、0 .75、1.0 0mg/L阿霉素 (ADR)的培养基培养转染细胞 48h后 ,采用3 H TdR掺入法检测细胞化疗药敏感性。结果 MG 63和空载体转染细胞增殖活性为 0 .197± 0 .0 15和 0 .195± 0 .0 2 1,差异无显著性 (P >0 .0 5 )。反义TGF β1转染细胞增殖活性为 0 .162± 0 .0 17,生长明显缓慢 ,对ADR敏感性明显增强 ,与对照组比较差异有显著性 (P <0 .0 5 )。讨论 通过阻断TGF β1自分泌环以降低骨肉瘤表达TGF β1,可以抑制细胞增殖活性 。 Objective To evaluate the effects of TGF-β1 autocrine blockage on the proliferation activity and drug sensitivity of osteosarcoma.Methods MTT method was used to detect proliferation activity of osteosarcoma cells transfected by antisense TGF-β1 gene.The transfected cells were cultured in medium containing adriamycin (concentration from 0 to 1.0 mg/L). 3H thymidine incorporation was used to investigate drug sensitivity.Results The proliferation activity of MG-63,MG-pcDNA3 and MG-TGF-β1 (-) was 0.197±0.015,0.195±0.021 and 0.162±0.017 respectively.The growth of MG-TGF-β1 (-) was suppressed markedly (P<0.05) and the sensitivity to ADR increased evidently (P<0.05).Conclusion To block TGF-β1 autocrine loop of osteosarcoma cells could inhibit cell proliferation and enhance sensitivity to chemotherapy drug.
