摘要
目的 研究 β 半乳糖苷酶 (β gal)在成骨细胞中的表达状况 ,为阐明MorquioB综合征的发病机制提供依据。方法 裸鼠各器官和骨组织标本行X gal染色检测。抽取羊和人骨髓行骨髓基质细胞 (BMSCs)培养 ,分为 4组 :Ⅰ :Adv hBMP 2转染组 ;Ⅱ :Adv β gal转染组 ;Ⅲ :未转染组 ;Ⅳ :地塞米松诱导组。分别行X gal染色和RT PCR检测 β gal的表达。 结果 裸鼠骺板两侧、骨膜内面及松质骨的成骨细胞和破骨细胞可见多量 β gal的表达。未转染BMSCs组有少量 β gal的表达 ,其他 3组细胞的 β gal表达增高。 结论 成骨细胞和破骨细胞可表达多量 β gal ,该两种细胞的β
Objective To study the status of β-galactosidase (β-gal) expression in osteoblast in vivo and in vitro,and try to clarify the pathogenesis of Morquio B syndrome.Methods Every organ and bone tissue of nude mice were stained with X-gal to detect the distribution of β-gal in vivo.The bone marrow stromal cells (BMSCs) of goats and human were cultured from bone marrow and divided into 4 groups: Ⅰ.Adv-hBMP-2 transduced group; Ⅱ.Adv-β-gal transduced group;Ⅲ.Untransduced group; Ⅳ.DXM induced group.β-gal expression of BMSCs was detected by X-gal staining and RT-PCR.Results The osteoblast and osteoclast beside the growth plate,or in the cambial layer of periosteum and cancellous bone,could express a large amount of β-gal.Untransduced BMSCs could express a little β-gal,while Adv-hBMP-2 and Adv-β-gal transduced BMSCs could express β-gal much more.Conlusion Osteoblast and osteoclast can express a large amount of β-gal.Maybe loss of β-gal of these cells is the immediate cause of Morquio B syndrome.
